Orphines
An emerging class of synthetic opioids. Background information from our friends at CFSRE. We have been tracking "cychlorphine" since our first detection in 2024, and had been expecting it to be among the broader class of orphines (synthetic opioids) to increase, but not displace fentanyl.
Naming of orphines is a mess. For example: N-propionitrile chlorphine = chlorphine = cychlorphine and "SR-17018" = 5,6-Dichloro Desmethylchlorphine. We are working with NIST to standardize naming, but be aware that this is in flux.
Pharmacology
N-propionitrile chlorphine = Cychlorphine, was tested at opioid receptors in Tango assays. It is a weak agonist at DOR and KOR, but it is a better agonist, more potent than DAMGO at MOR and than Nociceptin at NOP. Cychlorpine was screened at human GPCRome at final of 10 uM. In addition to opioid receptors, it also hits GPR97 and SSTR4. (via X-P Huang at UNC)

💡 Methods Considerations. We get more samples from certain states. Just because we haven't seen it in the lab, doesn't mean it can't be out there on there on the street. People preferentially send us samples when unexpected reactions happen. There is no such thing as a representative sample of the American drug supply.
